Description
Retatrutide (LY3437943) is a 39 amino acid single-molecule triple receptor agonist, supplied as a high-purity research peptide for laboratory and scientific use. Built on a GIP peptide backbone, it has concurrent activity at the GLP-1, GIP and glucagon (GCGR) receptors, with published binding data reporting EC50 values of 0.0643 nM at GIPR, 0.775 nM at GLP-1R and 5.79 nM at GCGR. Alongside the GLP-1R and GIPR mediated appetite and insulin signalling characteristic of the incretin class, the glucagon receptor arm is associated with increased energy expenditure, making retatrutide a reference compound for research into multi-receptor metabolic signalling, thermogenesis and adipose tissue biology. Clinical characterisation has progressed from the Phase 2 programme published in the New England Journal of Medicine (2023, n=338, 48 weeks) into the Phase 3 TRIUMPH programme, where TRIUMPH-1 (n=2,339, 80 weeks) reported dose-dependent mean body weight reduction of 19.0%, 25.9% and 28.3% at the 4mg, 9mg and 12mg doses respectively.
